Methodology

Computerized Imaging and Light Microscopy

Reflex Testing: When computerized imaging cannot be performed, testing is reflexed to light microscopy methodology only.

Performing Laboratory

Munson Medical Center Laboratories

Specimen Requirements

Submit only 1 of the following specimens:

Endocervical Brush/Spatula Collection Device

1. Obtain adequate specimen of ectocervix using a plastic spatula and of the endocervix using the cytobrush.

2. Rotate spatula over the squamo-columnar junction of the transformation zone 2 or more times.

3. Rinse spatula into the ThinPrep® PreservCyt® Solution vial by swirling vigorously in the vial 10 times.

4. Discard spatula.

5. Obtain specimen from endocervix using endocervical brush.

6. Insert brush in the cervix until only the bottomost fibers are exposed.

7. Slowly rotate 1/2 turn in 1 direction. Do not over-rotate.

8. Rinse brush into ThinPrep® PreservCyt® Solution vial by rotating the brush 10 times while pushing against the side of the vial.

9. Swirl brush vigorously as final step to further release material.

10. Discard the brush.

11. Tighten cap on vial so that the torque line on the cap passes the torque line on the vial.

12. Write patient’s name (first and last), specimen source, and date and time of collection.

13. Send specimen ambient.

Note: 1. For optimal interpretation, pap specimens should be collected near the middle of the menstrual cycle.

2. No douching or sexual intercourse for 24 hours prior to specimen collection.

3. Do not use lubrication jelly on the vaginal speculum. Tap water may be used as a lubricant when needed.

4. Mucus should be swabbed away prior to specimen collection.

5. The following information is required for processing:

A. Patient age or date of birth

B. Source (cervical, endocervical, or vaginal)

C. Name of ordering physician

D. Any pertinent clinical information

6. When computerized imaging cannot be performed, testing is reflexed to light microscopy methodology only.

Broom Collection Device Instructions

1. Obtain adequate specimen of cervix using the broom by inserting broom bristles into the endocervical canal deep enough to allow the shorter bristles to fully contact the ectocervix. Push gently and rotate the broom in a clockwise direction 5 times.

2. Rinse the broom into the ThinPrep® PreservCyt® Solution vial by pushing the broom into the bottom of the vial 10 times forcing the bristles apart. Swirl broom vigorously to further release material and then discard device.

3. Tighten cap on vial so that the torque line on the cap passes the torque line on the vial.

4. Write patient’s name (first and last), specimen source, and date and time of collection.
